Cell-permeable succinate improves amiodarone-induced mitochondrial Complex I and II inhibition in human platelets.

MitoEAGLE Short-Term Scientific Mission

Work Plan summary
1.Aim and motivation: explain the scientific and/or other motivation for the STSM and what outcomes you aim to accomplish with the STSM
Assessment of the bioenergetic profile of human peripheral blood cells is an emerging field of research with potential clinical applicability in the development of disease biomarkers. Early identification of mitochondrial dysfunction in mononuclear cells as biomarker in cardiometabolic diseases is currently highly investigated. mtDNA content might be an important biomarker in cardiovascular risk prediction and assessment of chronic cardiac diseases. Cellular metabolism plays a complex role in regulating immune processes. Immunometabolism research has focused mainly on macrophages that undergo metabolic reprogramming. Modulation of mitochondrial function in order to improve (re)polarization of inflammatory macrophages (M1) into anti-inflammatory(M2) is an emerging research field. Whether the permeable succinate might modulate this process it is not known.
The STSM objectives are to learn to perform high-resolution respirometry in cell lines as well as the techniques for mtDNA and citrate synthase assessment in blood cells.
2.Proposed contribution to the scientific objectives of the Action
This mobility will contribute to the scientific objectives of the Working Group 4 (MITO EAGLE data repository for blood cells). The techniques I will learn during this STSM will be further applied in the home institution during my doctoral studies and will allow us to carry on in the future parallel experiments and jointly publish the results under the umbrella of the MITO-EAGLE COST Action.
3. Techniques: pls detail what techniques or equipment you may learn to use
The techniques I will learn during this STSM are the assessment of mtDNA and citrate synthase activity. and high-resolution respirometry in blood cells by using the Oxygraph-2K equipment..
4. Planning- detail the steps you will take to achieve your proposed aim.
I gathered the background in using the HRR technique in assessing platelets respiration in my home institution and I will study the other related protocols. During the STSM I will participate in the experiments carried on in the host laboratory using the above mentioned techniques and then I will perform them myself.
