Ruthenium red

Description

Ruthenium red (synonym: ammoniated ruthenium oxychloride) inhibits the mitochondrial Ca2+ uniporter. However, in addition it has been shown to interact with and inhibit a large number of other proteins, including ion channels particularly of the Transient Receptor Potential Vanilloid (TRPV) family [1], Ca2+-ATPases, and, importantly, the voltage-dependent anion channel (VDAC) [2].

Abbreviation: RR

Application in HRR

RR: Ruthenium red (Ammoniated ruthenium oxychloride; (NH3)5RuORu(NH3)4ORu(NH3)5Cl6), Sigma-Aldrich: R2751, store at RT, CAS: 11103-72-3, M = 786.35 g·mol-1
Hazard statements: H302; Harmful if swallowed

Preparation of 2 mM stock solution (dissolved in H2O) for 2-mL O2k-chamber:
  1. Weigh 1.57 mg Ruthenium Red.
  2. Dissolve in 1 mL H2O.
  3. Divide into 0.1 mL aliquots.
  4. Store at -20 °C.

  • Titration volume (2-mL O2k-chamber): 1 µL using a 10 µL Hamilton syringe.
  • Final concentration: 1 µM.

Preparation of 0.5 mM stock solution (dissolved in H2O) for 0.5-mL O2k-chamber:
  1. Take 50 µL of the 1 mM stock solution.
  2. Add 150 µL H2O.
