MitoEAGLE Short-Term Scientific Mission

Work plan summary
1. Aim and motivation. Blood cells, like peripheral mononuclear cells (PBMCs) and platelets, can be obtained with minimal invasive procedures and used for analyzes of mitochondrial respiration for diagnostic and research purposes. Up to now I have analyzed by means of ELISA technique the expression of mitochondrial proteins in PBMCs from movement disorders and cancer patients. With the use of high-resolution respirometry I have analyzed mitochondrial respiration in PBMCs in movement disorders and cancer patients. The results were already published. Moreover, I have performed methodological study on the effects of anticoagulants on mitochondrial respiration in PBMCs. The results of the studies in movement disorders patients and on anticoagulant effects were my contributions to the Working Group 4 in COST Action MITOEAGLE. Currently, I do have the opportunity to perform analyzes of Ca+2 ions and mitochondrial membrane potential basing on tetraphenylphosphonium (TPP+) or TPMP+ evaluations using potentiometric ion-selective electrodes (ISE) and of reactive oxigen species using the system for high-resolution respirometry (HRR). However, ISE and HRR techniques require specific skills I would like to acquire during STSM I apply to and it is the aim of this STMS.
2. Proposed contributions to the scientific objectives of the Action I will contribute the results of the studies on calcium flow, mitochondrial membrane potential and production of reactive oxigen species in PBMCs in physiological and pathological conditions to the repository of Working Group 4 in COST Action MITOEAGLE. Moreover, I will continue the studies on methodology and protocols for mitochondrial analyzes in PBMCs using new techniques I will learn during STSM.
3. Techniques The techniques I will learn during STSM include potentiometric analyses using ion-selective electrodes (ISE) and high-resolution respirometry by means of MultiSensor applications. My training during STSM will fully contribute to the scientific objectives of MitoEAGLE according to the Memorandum of Understanding.
