Cyanide

Description

Potassium cyanide is a competitive inhibitor of cytochcrome c oxidase (CIV). Inhibition is reversed by pyruvate and high oxygen levels.

Abbreviation: Kcn

Reference: MiPNet09.12

MitoPedia topics: Inhibitor

Application in HRR

Kcn: Potassium cyanide (KCN); Fluka 60178, 100 g, store at R.T.; FW = 65.12

Caution: KCN is photosensitive, hygroscopic and very toxic! Prepare the solution fresh everyday!

Preparation of 1 M stock solution (dissolved H2O):

1) weigh 13 mg of Potassium cyanide
2) dissolve in 0.2 ml H2O
3) the pH of the solution may be very alkaline - adjust with HCl

Oxygraph-2k Manual Titrations: MiPNet09.12

  • Titration volume: 2 ยตl using a 10 ยตl syringe
  • Final conc. in 2 ml O2k-chamber: 1 mM
