Add catalase when measuring Complex IV (CIV) activity in permeabilized fibers (pfi)
Pfi have a high content of iron-containing proteins, which accelerate the autoxidation of tetramethyl-p-phenylenediamine (TMPD) in the CIV assay. Catalase prevents the accumulation of H2O2 generated by this autoxidation.

Do you observe simultaneous fluctuations in the O2 slope neg. and the Peltier power?
This can indicate fluctuations in the room temperature or electrical supply. Controlling the temperature or changing the electrical supply may resolve this problem.
