A first Advanced MultiSensor O2k-Workshop is organized for scientists who have participated previously in one of our introductory O2k-Courses. It will provide an overview on a variety of measurements in addition to O2 in the Oxygraph-2k. The two-chamber design of the O2k offers the possibility to perform simultaneous measurements either in each chamber equipped with ports for additional electrodes or light guides, or in parallel in two different chambers when using larger electrodes that do not fit through the PVDF stopper.
1.) TPP+ electrode and mitochondrial membrane potential, for meaurements with mitochondrial preparations (isolated mitochondria, permeabilized cells or permeabilized muscle fibres), and other ion selective electrodes.
2.) pH and metabolic acid production (aerobic or anaerobic glycolysis) in intact cells, including ph-stat applications.
3.) NO sensor for measurement of NO dynamics and mitochondrial oxygen kinetics (not in permeabilized tissue preparations with oxygen diffusion limitation).
4.) Oxystat: TIP-2k feedback control of oxygen levels and HRR; measurements at hypoxic steady-states.
5.) Spectrofluorimetry with wide range of applications: HRR and SAFAS Xenius spectrofluorimeter (isolated mitochondria, intact and permeabilized suspended cells).
6.) Spectrophotometry and cytochrome redox states: HRR and LEA Medizintechnik O2c (isolated mitochondria, intact and permeabilized suspended cells).
In addition to these MultiSensor topics, we will present and discuss (depending on specific interests of participants):
- DatLab 4.3 features for MultiSensor applications and analysis of steady-state experiments.
- Automatic instrumental background tests with dithionite titrations.
- Advanced protocols for respiratory OXPHOS control: multiple substrate-uncoupler-inhibitor titrations (SUIT protocols) and enzymatic thresholds.
- Oxygen kinetics obtained from aerobic-anaerobic transitions and respirometry at steady-state oxygen levels
